Objective: Measurement of circulating 25-hydroxyvitamin D3 is accepted as the clinical indicator of vitamin D status. It is very important of valid measurement of 25-hy-droxyvitamin D3 as clinical. Therefore there is need a standardization for measurement. Methods except tandem mass spectrometry (LC-MS/MS) measure total vitamin D and are less sensitive. LC-MS/MS is gold standard method for measuring vitamin D values. On this account we have invented a new method and evaluated this method to measure the imprecision and the accuracy and compared it with other systems. Materials and Methods: We used isotope-dilution LC-MS/MS (ID-LC-MS/MS) for determination of D3 with a stable-isotope-labeled internal standard. The values we obtained by ID-LC-MS/MS compared with HPLC, RIA and chemiluminescence methods. Results: The interassay CVs for 25-hydroxyvitamin D3 were 5.3%, 5.5% and 5.6% at 6.5, 24 and 43 ng/mL, respectively. The detection limit of the present method was <3.75 ng/ mL for 25-hydroxyvitamin D3. Total run time was 4 min. Method comparison between HPLC and LC-MS/MS showed a good correlation: y = 4.3822 + 1.0124 × (R2 = 0.96). Conclusions: We conclude that methods based on LC-MS/MS should be preferred in routine clinical work. We got the best results with HPLC according to the gold standard method, LC-MS/MS. © TurkJBiochem.com.
