Effects of in vitro acute hyperglycemia on lipid peroxidation in erythrocytes from diabetic patients and normal controls

Abstract

This study was aimed to determine whether the acute hyperglycemia causes erythrocyte membrane lipid peroxidation in patients with diabetes mellitus and in the normal healthy controls. Blood samples were obtained from type II diabetic patients, divided into two groups. One of them was used as diabetic group (Group I). Glucose was added to the remaining samples to form hyperglycemia and was called as glucose-pretreated diabetic group (Group Il). Blood samples taken from healthy subjects were divided into two groups. One of them was used as control group (Group III). The remaining samples were treated with D-glucose to form hyperglycemia and this group of samples was called as glucose-pretreated control group (Group IV). All samples were incubated with free radical producing agents, namely hydrogen peroxide, ferrous sulphate or xanthine/xanthine oxidase for two hours. Erythrocyte malondialdehyde (MDA) levels were measured before and after the incubation. Significant increase in MDA levels was recorded in the glucose-pretreated diabetic blood samples after period of the incubation (p<0.001). Similarly a remarkable increase in MDA levels was also evident in the diabetic blood samples incubated with xanthine/xanthine oxidase. Results of the present study indicate that the acute hyperglycemia may cause erythrocyte membrane lipid peroxidation in patients with diabetes but not in healthy controls. Under oxidative stress, acute hyperglycemia may also cause lipid peroxidation in the erythrocyte membrane of diabetic patients while no significant changes were observed in lipid peroxidation in the erythrocyte membrane of healthy controls.
